Currently the gap between desktop and service is getting smaller, if you are looking for creating a simple report connected to a semantic model online there is no major difference, the main differences currently is the user experience, and the fact that there is no on-object options.
Another difference is that you cannot interact with the model part in the same way you do in the desktop. In desktop you can create measures for that specific report, you can see the hidden columns and measures, you can save the the report has a Power BI Project.
So the main difference currently is the way you interact with the model part the rest of the creation for the report is very similar.
Also be aware that the new visuals that are showing up (new card visual, new slicer visual) are only available trough the preview features in the desktop version

Check out my blog: Power BI em PortuguêsThe biggest difference is that the user experience is simpler, faster and easier. For example, I cannot find the Optimization menu that I access via Power BI Desktop on the Service side.

The optimization menu is not available in the service, some of this options are part of the settings of the report other are not available in the service.
