If you mean all except on column you could do use the DAX measure ALLEXECPT
Otherwise if you are looking to filter out on particular value, you could use the FILTER measure

I created some data:
Add [amount1] and [amount2]
Here are the steps you can follow:
Create calculated column.
Sum_Column = 'Table'[amount1] + 'Table'[amount2]
Create measure.
Sum_Measure = SUM('Table'[amount1])+SUM('Table'[amount2])
