The Data Source Connection and Refresh Option is unavailable when I publish my PBIX file with MySQL connectivity in the Power BI online workspace. Any On-Premise Data Gateway connection is required for the MySQL server.

Hi. MySQL requires an On Premise Data Gateway. Once installed you need to configure its sources. Do you have any other source besides mysql? if it's onpremise you should configure that too, if it's cloud then we might need to talk about an option in the gateway to let you use hybrid sources (cloud and onpremise at the same time)
After configuring sources you can apply the Gateway Connection setting on the picture.

We launch a MySQL Server in an AWS EC2 instance. We Still need an On-Premise Data Gateway.
The Gateway is required for the source. It's not about cloud or onpremise. It's about MySQL requiring the gateway with the driver on the VM. You can install in the same EC2 or another EC2 with visibility of the server with the driver. Restart vm after installing the driver. You can test connection with PowerBi Desktop on that new gateway EC2 if you want. Once you have the gateway running in there, go to Power Bi Service and configure sources for the gateway.
