Is it possible to add calculations to an existing report in PowerBI service? or we can only add calculation to an existing report using powerBI desktop

Power BI Desktop is where you need to create the Measures.  Or create the Measures in the underlying Dataset, publish the Dataset, and the Live Connected Report using that Dataset will then have the Measures available to use within Service.
