I would like to add a column for calculation but I do not want to affect anything in the MySQL data source.
Does adding a new column works locally or it will sync to the source?
Do I need to have a new table for having a copy of data for the calculation?

and to add to Phil's post; you don't have to have a new table. The new calculated column can be ignored in terms of visuals you might build
however if the calculation involves a distinct value where the result is less records than the original table, such as a single sum for an overall category - it might be more useful to have a new stand alone table (you can join them)
and in either case you can undo these steps in your Query Editor should you need to experiment without affecting your core table

It definitely won't update your source data so you won't have to worry about that.
There is a tool that sounds like it might let you, but you'd need to install that separately.
