I have prepared a matrix table in PowerBI desktop which looks like the table below. However, each of the rows are measures e.g., net sales is a measure, staff salaries is a measure. I'd now like to add a column that calcuates the difference between the forecast and actual value. How can this be done?
To use each of the measures as rows I've used the "Switch Values to Rows" option.

Can you please share some dummy data of the table you have? Does it have a column listing "Actuals" and "Forecast" values? If "Actuals" and "Forecast" are from a column, I'm afraid this may not be possible in a Matrix visual. We may need to create a new table to help hold the measures. Without data I cannot give any detailed suggestions.
