I have a calculated column that looks like this:
Time to publish = DATEDIFF(related(Header[Quote Creation Timestamp]),QuoteActivity[Publish Live Timestamp], MINUTE)
I basically want to do this calculation but JUST for timestamps after 6:59am. I want to exlude rows in this calculation who's time stamp is before 6:59am.
Any help would be appreciated, thanks!

Use an IF clause in your formula. Something like this:
Time to publish = IF(QuoteActivity[Publish Live Timestamp] > TIME(6,59,0) , DATEDIFF(related(Header[Quote Creation Timestamp]),QuoteActivity[Publish Live Timestamp], MINUTE), BLANK())

As tested, tarunsingla's formula is correct when the QuoteActivity[Publish Live Timestamp] is of time type.
If QuoteActivity[Publish Live Timestamp] is in date/time format, go to Edit Queries->Add column->Time->only time, then use the time field in the "IF" function as tarunsingla's example.
