Is teher a way to add Start location and destination location slicer in Filled Map?

I don't think this is possible.

but, An idea about adding Map routing in Power BI has been submitted in the following link, please vote it up.
https://ideas.powerbi.com/forums/265200-power-bi-ideas/suggestions/8268978-power-bi-map-routing

Meanwhile, please check if the Flow map custom visual can satisfy your requirement.
