Hello,
I have 2 variables [Latitude] and [Longitude], and I would like to make a custom column which outputs in the following format: N[Latitude],E[Longitude] with "N" + "," + "E" being text.

The first solution is that you can simply use "&" symbol to concatenate thses columns and symbols in either DAX or Power Query as below:
Result = "N" & [Latitude] & "," & "E" & [Longitude]
Also, the second solution is that you can create a calculate column using DAX below:
Result = CONCATENATE ( "N", CONCATENATE ( [Latitude], CONCATENATE ( ",", CONCATENATE ( "E", [Longitude] ) ) ) )
