Diff Amount =
VAR _A =
    CALCULATE ( SUM ( 'YourTable'[Column 2] ), 'YourTable'[Column Name] = "A" )
VAR _B =
    CALCULATE ( SUM ( 'YourTable'[Column 2] ), 'YourTable'[Column Name] = "B" )
RETURN
    _B - _A
