I want to do the bold format of first row header column in matrix visual in stepped layout.
Can anyone help me on this?.

As far as I know, Power BI doesn't support specifying a column's font to be bold for the time being, you can only set all columns to be bold.
It is recommended that you apply conditional formatting to the background color and font color of the Value with an index of 1 to highlight the line or bold it to achieve a prominent effect.

If your data is of the following types, you may consider using the Specific column setting, but also can only change the color and not the font to bold:
You can submit an idea for it at Home (microsoft.com) and wait for users with the same needs as you to vote for you to help make it happen as soon as possible.
