Include AND Exclude in Calculate Measure

How do I add on to this to EXCLUDE values from columns?

TEST =
CALCULATE(
    COUNTA('EDW Issues'[Subject ID]),
     ALL('EDW Issues'),
    'EDW Issues'[IemRoleInCase] IN {"Subject", "Reporter"},
    'EDW Issues'[Title Categorized] IN {"Staff", "Management"})

    'EDW Issues'[Title Categorized] NOT IN {"XYZ", "ABC"}
)
 
consider using  || (or) , && (and) if you want to join multiple conditions.

NOT ( 'EDW Issues'[Title Categorized] IN {"XYZ", "ABC"} )

This did work after cleaning up few things in the data: 'EDW Issues'[IssueSub Formatted] <> "Ask a Question"

That gives me an error, unexpected expression...
