Currently that is not possible, you would have to open your PBIX, change the connection string.
Then upload to the Power BI Service.
You also would need to have the Power BI Gateway installed and configured with the data source to your On-Premise server.
And finally for your updated dataset you would have to change the connection to use the On-Premise connection.
