Count New =
CALCULATE (
COUNT ( 'Table'[Criteria] ),
FILTER ( 'Table', [Criteria] = "New" )
)Count Old =
CALCULATE (
COUNT ( 'Table'[Criteria] ),
FILTER ( 'Table', [Criteria] = "Old" )
)

I create some sample data for reference。
Then create the measure.
Count Measure =
CALCULATE (
COUNT ( 'Table'[Specimen] ),
FILTER ( 'Table', [Specimen] = "Specimen 1" || [Specimen] = "Specimen 2" )
)
The result is as follows.
