I would like to create a calculated column like below for the Matrix, is that possible?

For your requirement, I'm afraid that we should create measure to achieve your desired output.
Measure =
VAR day =
CALCULATE ( SUM ( 'Table'[Hours(Day)] ) )
VAR night =
CALCULATE ( SUM ( 'Table'[Hours(Night)] ) )
RETURN
DIVIDE ( day, day + night )
Here is the output. As you can see, if we create the calculated column, the total row will be incorrect.
