How to create measure for multiple values of a col...

I have a column in a table with multiple counts of same values. I want to create a measure with distinct values from this table and that measure will be used to calculate another measure. I can create a new column in another table for distinct value but that is serving the purpose.

I would appreciate if someone can provide any guideline.
