Current, we're on Power BI Report Server October 2020 version, and if we do an in-place upgrade of the server to the September 2021 version, would the old reports built using oct 2020 be still compatible on the upgraded server? In other words, can users still use Oct 2020 PBI Desktop developed reports and deploy to the upgraded server? Thank you in advance.

Yes. Users can still use Oct 2020 PBI Desktop developed reports and deploy to the upgraded server.
But it should be noted that some new features may not be able to use. For the updates of each version, you can check here:
