I have a Windows 2019 server that is hosting data for various companies.
How do I configure this web server so first of all it is accessible from PowerBI Desktop and Web data source?
Related to this is how do I ensure my clients only see their reports.
I will develop the Power BI reports for my clients and want to publish each clients' reports only for their use.

I am not 100% clear on all your questions but here is an attempt:
1. Once you install and configure your report server, you should be able to publish reports/models built using Power BI Desktop to the report server, as long as you are on the network.
2. To ensure your clients can only see their reports, you can create a folder for each client and setup security at the folder level. This will provide the client all the reports/content in that folder. See Secure Folders
3. If your clients are going to need to access the reports from outside the network, then you will need to configure Reporting Services for external access.
