In such situation, there are several useuful function.
If you want to calculate the difference of just houre or minute part if two Date.Time use
Duration.Minutes
Duration.Hours
Duration,Second
But if you want to calculate total differnce in hours or minute use
Duration.TotalHours
Duration.TotalMinutes

The result of datetime minus datetime is a duration type value. That's why you no longer see the datetime tools. If you want the result to be 12:47:05 AM, then you need to use Duration.TotalDays (not Duration.Days) and then Time.From. So
= Table.AddColumn(PriorStepOrTableName, "Time", each Time.From(Duration.TotalDays[Subtraction])))
