I have followed these steps to build a PowerBI report which use Excel Spreadsheet in the OneDrive for business as my data source.
Use OneDrive for Business links in Power BI Desktop
I see the data in my PowerBI report in PowerBI desktop. And when I publish the report to my workspace, I see the data in report as well. The issue is, when there are updates the Excel spreadsheet, I don't see the repot get updated with the latest data. And nothing happens when I click the refresh button in the report (the circle with the arrow).
I can only see the update when I manually go to Datasets + dataflows tab of My workspace, find the datasets of my report and click refresh.
My question is How can I setup my report so that when I click the refresh button in the power BI report, the report will get update with the latest data?

How did you deploy your report? Get data from a file that stored in onedrive and publish or upload your pbix file to it and get data from the power bi service?
If you mean the second scenario, power bi refresh only traces the pbix status(is not updated automatically), the underlying dataset update does not trigger the power bi refreshes.

Service refresh of sharepoint/onedrive excel files is supposed to happen every hour. You can also specify a schedule in the service if you need it more frequently.
NOTE: There is also the very exotic option of loading an excel workbook directly into the service. Not only can you use that as a direct query data source, Power BI service will as a bonus also update that Excel file's datasources for you.
Did you provide the one drive credentials in the datasource credentials section?

Thanks. Yes, I have entered my credentials in the datasource credentials section. I have used OAuth2.
And since it is an online excel spreadsheet, Import mode is my only choice in PowerBI desktop.
And the excel spreadsheet is small, it is only 128KB.
Moreover, in my PowerBI desktop, when I click Refresh, i see the visuals get update with changes in the spreadsheet. The problem is after I publish , Refresh in the report does not work.
