I have a dataset with column labeled "Status" with the values "Prospect" & "User". How do I create a measure to count all the prospects and users?

You can create one measure as below if you have similar table structure and data:
Countofuser = CALCULATE(COUNT('Users'[User Name]),filter('Users','Users'[Status]=SELECTEDVALUE('Users'[Status],"User")))
If the above methods are not applicable in your scenario, please provide the related table structure and more sample data(exclude sensitive data).
