1. Is it possible to add a new calculated measure (or) new calculated column to a table which is already published by me. If so anyone please let me know the procedure ?

1) You will need to export the .pbix (powerbi report file) that contains the data model, and edit that.
The procedure, once you have the file opened locally is:
2) As far as I know, if you use PowerBI dataset that has already been created and on the service as your datasource
ie using this option:
Then you cannot edit the dataset.
You will however, be able to edit the original using the steps outlined in 1)
I suggest you look into powerBI dataflows https://learn.microsoft.com/en-us/power-bi/transform-model/dataflows/dataflows-introduction-self-ser... this might fit your situation better.
